What is GSB-214?

GSB-214 (ГСБ-214) is a synthetic dimeric dipeptide designed to mimic loop 1 of BDNF: bis(N-monosuccinyl-L-methionyl-L-serine) heptamethylenediamide. It is a distinct compound from GSB-106, which was designed around a different BDNF loop. The Russian synthesis paper dates to 2012. Sharing a design concept does not make their experimental results or safety data interchangeable. C01 (opens in a new tab)

Cell signaling is not a binding-affinity measurement

A 2016 study measured increased TrkB and AKT phosphorylation in mouse hippocampal cells exposed to GSB-214. ERK activation was not detected at the tested concentration and time points. This supports a particular signaling pattern under those conditions; it does not establish an exclusive mechanism in every tissue. These assays did not determine a direct receptor-binding affinity. The BDNF–TrkB guide explains the endogenous pathway separately. C02 (opens in a new tab)

Section 3 of 8Stroke-model benefits did not cover every outcome

Stroke-model benefits did not cover every outcome

In the same rat study, intraperitoneal GSB-214 treatment began after experimentally induced ischemia. Infarct volume was approximately 28% lower than in the ischemic control group, and limb-placing performance improved. The cylinder test of forelimb-use asymmetry showed no statistically significant benefit. The GSB-214 group contained only 6 rats. Methods and results place infarct measurement at day 21, while the figure caption says day 7; this inconsistency remains unresolved. These findings do not establish benefit after stroke in humans. Section 4 of 8Memory effects depended on the model and dose

Memory effects depended on the model and dose

A 2022 study in male rats used novel-object recognition after chemically induced memory impairment. In the scopolamine model, the lower tested doses improved long-term recognition, but no significant short-term benefit was established; the highest tested dose was inactive. In the streptozotocin model, short-term recognition improved, while the model itself produced no significant long-term deficit. These are different outcomes in experimental models, not evidence of treating Alzheimer’s disease in patients. The Russian and English versions report the same study and must not be counted as separate replications. C04 (opens in a new tab)

What does the receptor-blocker experiment show?

In a separate experiment within that paper, the Trk inhibitor K252A abolished the long-term recognition benefit of GSB-214 in rats without the chemically induced disease models. This supports involvement of Trk signaling. It is not a direct binding measurement, does not by itself distinguish every Trk receptor, and does not demonstrate the same mechanism in patients. The inhibitor experiment must not be described as if it had been performed within both disease models. C05 (opens in a new tab)

Section 6 of 8A single dose and repeated treatment gave different findings

A single dose and repeated treatment gave different findings

In a 2022 mouse forced-swim study, a single administration of GSB-214 showed no antidepressant-like activity at the tested dose. After five daily administrations, immobility was reduced by 18.2% versus control. This qualifies the older negative acute findings: the effect depended on the experimental schedule. Reduced immobility is a behavioral test result, not a diagnosis of recovery from depression or evidence of a human treatment schedule. C06 (opens in a new tab)

Section 7 of 8Metabolic findings remain preclinical

Metabolic findings remain preclinical

The indexed abstract of a 2020 mouse diabetes study reports reduced hyperglycemia and prevention of weight loss; a PI3K inhibitor abolished these effects. A 2022 study abstract reports improvements in oxidative-stress markers, but inhibition attenuated the glutathione-peroxidase effect and not the malondialdehyde effect. Only abstracts were reviewed for these two papers. Different endpoints need not share an identical mechanism, and these findings do not establish diabetes treatment in people. What remains unknown about human use?

The reviewed cell and animal studies do not establish clinical efficacy, a human adverse-event profile, human pharmacokinetics or a validated human dose. Efficacy experiments are not a comprehensive toxicology program. Several papers come from overlapping research teams; multiple publications do not automatically mean independent replication. Data from related mimetics must not fill these gaps. This focused review is not an exhaustive clinical-trial registry search. C08 (opens in a new tab)
