What is GK-2, and why does the suffix matter?

GK-2 (ГК-2) is a synthetic dimeric dipeptide mimetic designed from a loop of rat nerve growth factor: bis(N-succinyl-L-glutamyl-L-lysine) hexamethylenediamide. GK-2(h) contains glycine instead of glutamic acid and was designed from the corresponding human NGF loop. They are different compounds; findings must not be pooled under one name. Neither is the full NGF protein. The NGF–TrkA guide explains the endogenous system. C01 (opens in a new tab)

Section 2 of 8What does the mechanism study support?

What does the mechanism study support?

In a 2015 cell study, GK-2 increased TrkA and AKT phosphorylation, while ERK activation was not detected at the tested time points. A PI3K inhibitor abolished the measured protection against oxidative injury. GK-2 did not induce neurite outgrowth in the tested PC12 assay. These results support a particular signaling pattern under those conditions; they do not show that GK-2 reproduces every NGF effect or prove direct receptor binding from phosphorylation alone. Section 3 of 8What improved in the memory experiments?

What improved in the memory experiments?

A 2013 study reported improved habituation and spatial-memory performance in two rat injury models used to study aspects of Alzheimer’s disease. GK-2 was given intraperitoneally after the experimental injury. The groups were small, and these induced models do not reproduce the full human disease. The findings concern performance in those animal tests, not demonstrated prevention or treatment of Alzheimer’s disease in people. C03 (opens in a new tab)

Section 4 of 8Positive markers and results that did not reach significance

Positive markers and results that did not reach significance

In the 2019 rat ischemia study, GK-2 increased the hippocampal proliferation marker Ki67. The striatal Ki67 result was a trend (p = 0.08). The postsynaptic marker PSD-95 improved when administration began after 6 hours; after 24 hours its change was only a trend (p = 0.08). No significant effect was found on synaptophysin. These markers do not by themselves demonstrate new functioning neurons or restored neural connections. The authors’ interpretation of neuroregeneration is broader than the direct measurements. Section 5 of 8Does the mouse behavior result depend on duration?

Does the mouse behavior result depend on duration?

In a 2022 forced-swim study in mice, acute GK-2 administration did not significantly reduce immobility. After 5 days, one tested dose reduced immobility by 13.2%. This is an antidepressant-like screening result, not evidence that GK-2 treats human depression. The acute null result and the repeated-administration result must both remain visible. Section 6 of 8What do the safety experiments actually establish?

What do the safety experiments actually establish?

The 2015 rat experiments detected no increased pain sensitivity in the tested tail-withdrawal assay and no body-weight effect after 14 days of intraperitoneal GK-2. This is a favorable finding for those endpoints, not proof of an absence of side effects. It does not establish long-term human safety, reproductive safety, interactions or a safe human dose. The reviewed evidence does not provide a verified human adverse-event profile. C06 (opens in a new tab)

Section 7 of 8What does the newer Russian mouse study add?

What does the newer Russian mouse study add?

The Russian abstract of a 2024 study describes mice selected for large or small relative brain weight. The authors report genotype-dependent behavioral tendencies but explicitly state that small groups prevented detection of significant differences from controls in the cognitive test. They also report a more active response to novel food. Only the abstract was reviewed here. This study cannot be presented as a confirmed improvement in cognition. C07 (opens in a new tab)

Section 8 of 8What is new in the project report, and what remains unknown?

What is new in the project report, and what remains unknown?

A Russian Science Foundation project reports comparative cell experiments from 2024 and 2025 on GK-2 and GK-2(h), including differences in signaling between mouse hippocampal and human neuroblastoma cell lines. Its later results include PLCγ activation, so “only AKT” should not be used as a timeless description. This is an investigator-authored project report, not a human clinical trial or an independently replicated result. Human-derived tumor cell lines are not human participants. Clinical benefit and a usable human safety profile remain unestablished by these sources. C08 (opens in a new tab)
